The changes in gene expression were analyzed in Fc(aMD4)B3-treated and PBS-treated PXB-mice livers. Fc(aMD4)B3 is a surrogate c-Met receptor agonist made by lasso-grafting c-Met-binding macrocyclic peptide aMD4 into B3 position of Fc. The in vivo activity of Fc(aMD4)B3 was examined using chimeric mice transplanted with human hepatocytes (PXB-mice). PXB-mice (PhoenixBio) were generated from urokinase-type plasminogen activator-cDNA/severe combined immunodeficiency mice transplanted with human hepatocytes with approximately 80% of hepatocytes being humanized. PXB-mice (male, > 12 weeks, 15 to 21 g) were given a single subcutaneous injection at 5 mg/kg for Fc(aMD4)B3 in 200 ul PBS or control PBS. Forty-six hours after administration of Fc(aMD4)B3 or control PBS, livers were isolated from PXB-mice and treated with RNAlater for RNA-Seq.
