PGC-1a overexpression in neurons protects against chronic cerebral hypoperfusion-induced cognitive impairment in mice. To investigate the neuroprotective mechanism of PGC-1a, we employed RNA-Seq analysis on PGC-1a-overexpressed HT-22 cells, a hippocampal neuron cell line. We performed OGD experiment to mimick chronic cerebral hypoperfusion. Indeed, PGC-1a overexpression alters the gene expression profiles of HT-22 cells, suggesting that neuronal PGC-1a might play an important role after chronic cerebral hypoperfusion. Overall design: PGC-1a-overexpressed HT-22 cells and control were subjected to the hypoxia incubator chamber (5% CO2 and 95% N2) and exposed to oxygen and glucose deprivation (OGD) treatment for 2 h in EMEM medium without serum/glucose. After that, HT-22 cells were cultured in normal medium at 37°C for 24 h. Then the samples were processed to perform RNA-Seq analysis.
