TGF-b-dependent Lymphoid Tissue Residency of Stem-like T cells Limits the Response to Tumor Vaccine

We report the transcriptional profiles of tumor-specific CD8+ T cells isolated from tumor draining lymph nodes (TDLN) and tumor following tumor vaccination in C57BL/6 mice. In addition, we included wild type (WT) and TGF-b receptor conditional knockout (Tgfbr2-/-) CD8+ T cells. Overall design: WT and Tgfbr2-/- Pmel-1 TCR transgenic T cells were adoptively transferred into recipient mice followed by B16 tumor subcutaneous inoculation. Around 14 days later, all recipient mice were immunized with Pmel-1 cognate peptide plus poly I:C. Together with tumor vaccination, all mice received FTY720 treatment to limit cell migration. Donor Pmel-1 T cells were FACS sorted 4 days after vaccination and subjected to RNA-seq analysis.
