In this study, mouse macrophage Raw264.7 was pretreated with Lespedeza bicolor root components and then stimulated with LPS and then high-throughput sequencing technology was used to explore the expression of inflammation-related genes of Raw264.7 in mouse macrophages treated with Lespedeza bicolor root components, and to find interesting targets,NF-kappaB and p38 signaling pathway. RAW 264.7 cells were seeded in 100 mm dishes at a density of 1 × 107 cells per well.Three experimental groups, Control, LPS, and LB+ LPS, were set up, and 0.5 μg/mL LPS was added to stimulate for 6 h after LB was activated for 30 min. After treatment, the cells were collected, and total RNA from each group was extracted using TRIzol (TaKaRa, Japan). Paired-end sequencing was ultimately carried out on an Illumina HiSeq 4000 platform at LC-BIO Technologies (Hangzhou, China).
