To gain further insight into the possible mechanism(s) by which S1P regulates cell signaling pathways in human myometrial cells, RNAseq was performed using two immortalized myometrial cell lines following treatment with exogenous S1P or vehicle control for 3 h. Overall design: Two immortalized human myometrial cell lines were used in the experiments. hTERT-HM (hHM) is an hTERT-immortalized myometrial cell line obtained from a non-pregnant human uterus at the time of hysterectomy and were a kind gift from Dr. Jennifer Condon. The second cell line, HM6ERM2 (HM6), was previously derived from term-pregnant human uterine tissue and immortalized using hTERT as previously described. Cells were treated with either S1P or vehicle control ( bovine serum albumin). Each treatment was performed in triplicate for each of the two cell lines.
