Overall, this study generated NANOS3 knockout cattle via CRISPR/Cas9-mediated zygote editing and characterized their germline development from fetal through reproductive age. Data has been deposited in NCBI’s Sequence Read Archive (SRA) repository and the NCBI Gene Expression Omnibus (GEO) repository. Accession: PRJNA1026125 (SAMN37734900–SAMN37734907): PacBio long-read amplicon sequencing of bovine NANOS3. This study employed cytoplasmic microinjection of the CRISPR/Cas9 system in in vitro-produced bovine zygotes to generate NANOS3 knockout bovine samples. Long-read amplicon sequencing was used to evaluate the resulting mutations present in each sample. GSE244321: We performed single cell RNA-sequencing (scRNA-Seq) of testes from bovine fetuses derived from either CRISPR/Cas9 NANOS3 knockout (KO; NANOS3 -/-) or wildtype control (NANOS3 +/+) embryos. The scRNA-Seq analysis showed a complete loss of primordial germ cells (PGCs) and gonocytes in NANOS3 KO fetal testes, while maintaining the development of somatic support cells.
