Pediatric GLP-1 Receptor Agonist Adverse Event Phenotype Framework: Preliminary Terminology and Candidate Mappings (Version 1.0)

Overview This repository provides a preliminary, structured phenotype framework for identifying and categorizing potential adverse events associated with glucagon-like peptide-1 receptor agonists (GLP-1RAs) in pediatric and young adult populations younger than 21 years. The framework comprises 52 prespecified adverse-event phenotypes organized into five clinical domains: (1) gastrointestinal, (2) metabolic and nutritional, (3) neuropsychiatric, (4) systemic, renal, and other, and (5) endocrine outcomes. Resource Description The repository provides a structured phenotype classification table with candidate ICD-10-CM codes and biomedical ontology annotations. The framework was developed to support standardized outcome definitions, terminology harmonization, and reproducible research involving electronic health records, administrative claims, and other real-world data sources. Potential Applications This resource may support: Development of candidate outcome definitions for pediatric GLP-1RA safety research. Harmonization of adverse-event phenotypes across observational data sources. Design of pharmacoepidemiologic studies using administrative claims and electronic health records.
