The equilibrium binding constant of 4M5.3 scFv binding to the epitope conjugate, fluorescein-biotin, was determined by a competition assay of 4M5.3 displayed on the surface of yeast with soluble 4-4-20 scFv. The affinity matured 4M5.3 scFv has a 1800-fold higher affinity than the parent 4-4-20 antibody (KD = 0.7 nM).
