This is a modified version of a Drosophila expression construct called JFRC14 (Pfeiffer et al. 2010). A new restriction site (SpeI) was created upstream of the codon-optimzed GFP coding sequence so that any cDNA of interest can be subcloned in-frame with GFP. This allows robust expression levels of GFP-fusion proteins in the presence of a Gal4 driver. Briefly, the original JFRC14 plasmid was digested with KpnI and EcoR1 to release the GFP-WPRE-SV40 cassette, and in its place a [KpnI-cut]-{SpeI}-GFP-WPRE-SV40-(EcoRI-cut) ~2Kb-PCR-product was introduced. The PCR primers were: Fwd:5'-GAGA[GGTACC]AATCAAAATG{ACTAGT}TCCAAAGGTGAAGAACTGTTTACC-3' and Rev:5'-TTCAA(GAATTC)GTTTAAACGGCCGGCCAGATCGATC-3'.
