Human CD34+ hematopoietic stem and progenitor cell expressing MLL-Af4 phenocopies t(4;11) pro-B acute lymphoblastic leukemia In Vivo. A Novel Human MLL-AF4 Model

We assembled MLL-murine Af4 retroviruse and transduced human CD34+ cells, which can initiate ALL upon transplantation into NSG mice. The RNAseq was performed MLL-Af4 CD19+CD34+ leukemic cells from NSG mouse bone marrow to determine the fidelity of expression profile of the model to patient samples The leukemia initiated by MLL-AF9 and normal human proB cells engrafted in NSG mice were used for RNAseq as control.
