Evolve and resequence study

Illumina sequencing libraries were made directly from genomic DNA of clones and population evolved under sulfate-limited conditions into the chemostats and stored in glycerol at -80C. Genomic DNA was extracted from dry, frozen cell pellets for the population or individual colonies using the Smash-and-Grab method (Hoffman and Winston 1987). Genomic DNA libraries were prepared for Illumina sequencing using the Nextera sample prep kit (Illumina). Barcoded libraries were quantified on an Invitrogen Qubit Fluorometer and submitted for 75 bp paired end sequencing on an Illumina HiSeq 2000 or a MiSeq.
