By using luminal lineage tracing model, here we report the regulatory network underlying intrinsically castration-resistant LY6D+ luminal progenitors during prostate tumourigenesis. The luminal cell-specific PTEN deficient mouse model of PCa (K8-CreERT2;PtenFlox/Flox;R26-StopFlox/Flox-EYFP (K8PY) and the control K8-CreERT2;R26-StopFlox/Flox-EYFP (K8Y) mice) are used in this study. Overall design: K8PY and K8Y mice (>8 weeks old) were treated with tamoxifen to induce Pten deletion and/or YFP expression in the K8+ luminal cells, either hormone sensitive mice or one-month post-surgical castration. Four months post-tamoxifen, primary prostate cells were isolated and luminal prostate cells were FACS-sorted by YFP+LY6D+ and YFP+LY6D- from K8Y and K8PY mice. Total RNA from FACS-purified luminal cells was isolated using an RNA/DNA Purification Micro Kit (Norgen, #50300)
