Metformin-responsive plasma extracellular vesicle miRNA profiles in polycystic ovary syndrome with insulin resistance

This project investigates how metformin therapy alters plasma extracellular vesicle (EV)-associated microRNAs in women with polycystic ovary syndrome and insulin resistance (PCOS-IR) and how these changes may relate to ovarian regulation. EV-associated small RNAs were sequenced from plasma samples collected from six women with PCOS-IR before and after 3-month metformin treatment (paired design), and from six healthy controls. EVs were isolated from plasma using a standardised ultracentrifugation-based protocol, and small RNA libraries were prepared with a dedicated miRNA kit and sequenced on an Illumina NovaSeq 6000 platform. The resulting dataset includes raw small RNA sequencing reads suitable for downstream miRNA expression profiling, differential expression and pathway analyses.
