Single-embryo RNA-seq analysis in LAD disrupted 2-cell embryos.

We investigated whether LAD disruption phenotypes are associated with transcriptional changes during zygotic genome activation (ZGA). To address this, we performed single-embryo RNA sequencing at the late 2-cell stage and analyzed the transcriptome. Overall design: Mouse embryos (zygotes) were microinjected with mRNA(s) encoding either mGFP alone (Control) or candidate proteins from the respective pools. The embryos were cultured until the late 2-cell stage and collected for Smart-Seq+5' analysis. Embryos were processed in two batches: batch 1 (collections 1 and 2) included mGFP, Pool J, and Pool M samples, while batch 2 (collections 3 and 4) consisted of Pool I, Pool N, and an additional set of mGFP embryos as controls.
