Broilers supply a significant quantity of high-quality chicken meat to humans and enhance their level of life. Better understanding the exceptional qualities of broiler breeders and increasing breeding efficiency requires an appreciation of the variety of local livestock and poultry. Sichuan province's Pingwu Red chicken is a good native poultry genetic resource. The identification of DNA polymorphisms within the Pingwu Red chicken genome falls behind those of conventional broiler breeds. To reveal genetic variety, blood genomic DNA was extracted from Pingwu Red chickens and processed for SNP marker collection and phylogenetic tree building using SLAF-seq. In this study, the genetic background linkages of 18 Pingwu Red chickens were studied using high-throughput locus-specific amplified fragment sequencing (SLAF-seq) to find meaningful single nucleotide polymorphisms (SNP) across all accessions. The collected 952,944 high quality SNPs (integrity >0.5 and MAF >0.05) were utilised to explore the genetic evolutionary connection of Pingwu Red chickens. The phylogenetic tree genetic study effectively revealed the genetic basis of Pingwu Red chicken, revealing that all samples belonged to the same genetic group. Specific length amplified fragment sequencing (SLAF-seq) is a high-resolution method for de novo SNP identification and genotyping. In conclusion, the research demonstrates that slaf-seq is an effective tool for revealing the genetic history of Pingwu red chicken, providing a scientific foundation for the use of Pingwu red chicken in broiler breed selection.
