Suppression of mosquito Aedes aegypti population using a CRISPR-targeted leucine aminopeptidase 1 mutation

The spermathecae from 2000 individual mosquitoes at the virgin, 72 h PE, and 5 days PBM stages were pooled for the construction of a spectral library. In addition, the spermathecae from 500 individual mosquitoes at the same time point as described above were collected for sequencing. Then, sequencing was performed by Beijing Protein Innovation Co., Ltd. In brief, the protein is first extracted, enzymatically digested, and purified. The enzymatically digested peptides are then subjected to a shotgun assay, followed by a database search to obtain protein characterization results. Based on the shotgun results, mass spectrometry parameters such as the scan interval of the DIA are determined and the sample is subjected to DIA. The results of the shotgun search were used to build a library of ion spectra (ion library) and the detected spectra of the DIA samples were matched and quantitative information was extracted from the ion library using Peakview. Finally, the protein results from Peakview were statistically analyzed to obtain the results of the analysis of variance.
