KLF1 is to regulate the gamma-globin to beta-globin gene switch during erythropoiesis. The goal of this study was to discover genome-wide transcriptome expression pattern and identify direct targets of KLF1 downregulation involved in gamma-globin related to transcription induction. To determine the genome-wide transcriptome expression pattern during erythropoiesis under KLF1 silencing, we applied for a short hairpin lentiviral vector carrying a puromycin resistance gene with shKLF1 transduced into normal erythroid progenitors. At day-15 in the 28-day culture period, at day-15 we found that KLF1 was silenced so that we used the RNA from cells to run RNA-seq.
