This repository contains single-cell RNA and single-cell ATAC sequencing data of peripheral blood mononuclear cells (PBMCs) from melanoma patients at baseline (prior to receiving anti-PD1 therapy) and healthy donors. Patients were selected based on their IFN-I response capacity (measured by mass cytometry), which is associated with patient survival after therapy. The purpose of the experiment was to better understand the mechanisms underlying the differences in outcome observed betwee the two patient groups. 8 patient PBMCs (4 with high IFN-I response capacity (IRC) and 4 with low IRC) and 2 healthy donor PBMCs were sequenced through multiome (scRNA + scATAC) sequencing. Briefly, cryopreseved PBMCs were thawed and 10X multiome seuqencing workflow was performed following DNAse treatment according to the manufacturer's protocol. Processing and sequencing was performed by the Princess Margaret Cancer Center Genomics Core.
