Genome cytosine methylation among isolates of Calonectria pseudonaviculata (Cps) with differential sporulation during boxwood infection

10-12 days old mycelia culture grown in potato dextrose broth of 8 Cps isolates with differential sporulation capacities were sequenced on MinION with Oxford Nanopore Sequencing Technology modified basecalling model. The analysis of cytosine methylation of these isolates showed that isolates with a poor sporulation capacity had higher levels of 5mC in hypothetical zinc-finger domain protein and non-ribosomal peptide synthetase genes than the great conidial producers. This finding suggests functions of these genes associated with sporulation and diversity of Cps regulated by methylation, providing insights into mechanisms underlying the transmissibility and pathogenicity of Cps.
