After one week of adaptive feeding, 8-week-old male C57BL/6J mice were randomly divided into five groups using a random number table: control (Con) group, CDDP group, low-dose Gen (Gen-L) group, medium dose Gen (Gen-M) group, and high-dose Gen (Gen-H) group, with 8 mice in each group. Based on the preliminary experiments, the doses of Gen were 25, 50, and 100 mg/kg, administered orally once a day for 7 consecutive days. Subsequently, except for the Con group, all other groups of mice were established with AKI models by a single intraperitoneal injection of CDDP (20 mg/kg), while the Con group was injected with an equal volume of physiological saline. During the entire administration and modeling period, mice that did not meet the criteria were excluded based on the following criteria: ① Death during administration; ② Modeling failure; ③ Other mice that cannot be further observed. Ultimately, the modeling success rate was 85%. The basis for successful modeling includes: ① Clinical symptoms: typical symptoms such as reduced activity and weight loss in mice within 24 hours after CDDP injection; ② Biochemical indicators: After 48 hours of modeling, the levels of SCr and BUN significantly increased; ③ Renal injury markers: After modeling, the levels of renal injury factors KIM-1 and NGAL significantly increased; ④ Pathological changes: Typical pathological changes such as swelling, necrosis, and inflammatory cell infiltration of renal tubular epithelial cells appear. By detecting urea nitrogen and creatinine, kidney injury molecule-1 (KIM-1), and neutrophil gelatinase associated lipocalin (NGAL), combined with HE, Masson, PAS staining, renal function and pathological damage were evaluated, and inflammatory response and oxidative stress related indicators were measured.
