Here, we used ADMSC overexpressing SPARC to treat dry eye induced by benzalkonium chloride in dogs. The efficacy of SPARC-modified ADMSC for dry eye was verified by detecting ocular surface inflammation indices, corneal fluorescein sodium staining, basal tear secretion, and histopathological observations. In vitro validation was also performed using benzalkonium chloride-induced HCECs damage. The rescue effect of corneal epithelial cells on apoptosis after addition of SPARC-modified ADMSC supernatant in culture by cell migration, Giemsa staining, CCK8, flow cytometry, immunofluorescence. Analysis of the transcriptome sequencing data indicated that SPARC was designed to improve its therapeutic effect on dry eye by enhancing the in vitro viability, proliferation, immunosuppression, vascular regeneration and anti-scarring, which in turn promotes the repair of corneal epithelial cells, modulates the production of inflammatory mediators, and improves its therapeutic effect on dry eye syndrome. This study offers the most current treatment strategy and theoretical foundation for addressing dry eye.
