DmPGE2 predominantly interferes with IR-induced p53-downstream apoptotic gene expression at 1 h, and suppresses IR-induced cell cycle gene expression at 24 h. Overall design: Fgd5 mice (C57BL/6 background with ZsGreen fluorescent HSC) were treated with dmPGE2 or vehicle (8 mice/group) 30 min prior to irradiation at the 50% lethal dose of 8.53 Gy. Non-irradiated control groups were also treated with dmPGE2 or vehicle (7-8 mice/group). At 1 h and 24 h post-irradiation, alongside treated non-irradiated controls, bone marrow cells were extracted and HSC sorted for RNA sequencing by SLAM-LSK(wide) gating and Fgd5/ZsGreen fluorescence as described.
