Mini-dataset on the expression of the nirA gene from the cyanobacterium Anabaena in individual cells

Description This dataset provides single-cell experimental data to investigate the role of intrinsic noise on gene expression in the multicellular cyanobacterium Anabaena sp. PCC 7120. The primary objective is to evaluate whether cell size and morphological variables (potential sources of biological noise) influence the transcriptional activity of the PnirApromoter. Methodological Summary Biological System: A reporter strain carrying a transcriptional fusion integrated into the alpha-plasmid neutral site (nucA/nuiA). Data Acquisition: Three-channel confocal microscopy (mVenus/EYFP, Autofluorescence, and Brightfield) using an Olympus FLUOVIEW FV3000. Image Processing: Automated cell segmentation performed on the red autofluorescence channel using a custom ImageJ/Fiji pipeline. Data Curation: The dataset includes raw measurements (v0.1) and a curated version (v0.2) filtered by cell area (3-15 µm²) to ensure data quality. Key Findings & Contents Statistical Analysis: Principal Component Analysis (PCA) was conducted to identify latent correlations between morphology and expression. Dataset Files: Includes raw .tif micrographs, ImageJ ROI masks (.zip), the processing script (.txt), and curated data in .csv format.
