Provided are a rice fertility regulatory gene and a mutant and the use thereof. Provided is a rice gene GMS1 with the functions of regulating male germ cell development in rice and of pollen fertility， wherein the CDS sequence thereof is as shown in SEQ ID NO: 2， and the amino acid sequence is as shown in SEQ ID NO: 3. Provided are a radiation mutagenesis mutant and a CRISPR-knockout mutant of the GMS1 gene， and also provided is a molecular marker identification method of the mutants. The rice gene GMS1 provided can be used in sterile seed production and the manufacturing of rice hybrids， thus possessing both great application value and economic value.
