Supporting data for “Discovery of a Covalent Ligand of Pyruvate Kinase M2 for Treating Hepatocellular Carcinoma”

The research is about developing a covalent drug target pyruvate kinase M2 to treat hepatocellular carcinoma. Activating PKM2 by promoting tetramer formation were found to be anti-tumour. We have identified 2J1, a cysteine targeting compound, was able to bind and activate PKM2. We have used gel-based ABPP, pyruvate kinase activity assay, RNA-seq, cellular fractionation, flow cytometry for cell cycle analysis, confocal imaging for ROS measurement, metabolomics, and mouse experiment to investigate 2J1 effects in PKM2 and HCC cells. This data set contains western images for cellular fractionation, cell cycle marker and Flag-pull down. It also includes rhodamine gel images for ABPP, and omics data such as proteomics, metabolomics and transcriptomic. It contains data analysis for in vivo mouse experiment as well.
