We imposed the salinity stress by superimposing the standard Hoagland solution with gradually increasing NaCl concentration in the following manner: 50 mM for two days, 75 mM for two days, 100 mM for two days, and 125 mM for two days. Finally, after exposure to gradually increasing salinity for eight days, the seedlings were subjected to a salinity stress of 150 mM NaCl. The samples were collected after 6 hours, 12 hours, and 24 hours after stress imposition from both control and salt-stressed plants. RNA was isolated from the individual samples and a salt stress RNA pool and a control RNA pool were prepared by taking the equimolar amount of RNAs from each time point of stress. Both the control RNA pool and the salt stress RNA pool were used for RNASeq sequencing.
