This project aims to investigate whether glycan modifications influence the transcriptome of HCT116 cells, a human colorectal carcinoma cell line with a fucosylation deficiency caused by a mutation in GDP-mannose-4,6-dehydratase (GMDS). Using glycan modifiers such as fucose, 3-fluorinated sialic acid (3-F-Sia), and Kifunensine (Kif), we generated five distinct glycan phenotypes: sialylated (S), Underacted(Underc), fucosylated (F), sialofucosylated (FS), and high mannose (HM). mRNA sequencing will be performed to explore whether these glycan modifications induce significant transcriptomic changes
