In a TBX5-linked human cellular model of atrial disease, we examined the gene networks controlled by TBX5 during human atrial differentiation using single-cell RNA and ATAC sequencing. Overall design: Wildtype, TBX5 heterozygous and homozygous mutant iPSCs were differentiated to atrial cardiomyocytes. At day 20 of differentiation, cells were harvested for single-cell ATAC sequencing. Two replicates per genotype per timepoint were harvested. Wildtype iPSCs were also differentiated to ventricular cardiomyocytes and cells were harvested at day 23 for single-cell ATAC sequencing.
