We applied LEAD-m6A-seq to 11 sites of HeLa mRNA to probe m6A modification at the target sites. Overall design: DNA probes were annealed to mRNA extracted from HeLa cells. The annealing product underwent cDNA extension reaction mediated by Bst DNA polymerase and was then subjected to library construction and illumina sequencing.
