HSF1–MYCN axis, a transcription-dependent regulatory axis, could be a prognostic indicator and potential therapeutic target in liver cancer. Overall design: Identify downstream target genes co-regulated by HSF1 and MYCN in Huh7 cells using whole transcriptome sequencing (RNA-Seq). Investigate the binding sites of HSF1 and H3K27ac in Huh7 cells using Cleavage Under Targets and Tagmentation (CUT&Tag) with specific antibodies. Study the H3K27ac status and enhancer changes influenced by HSF1 in Huh7 cells using chromatin immunoprecipitation and sequencing (ChIP-seq) with specific antibodies.
