The myometrium undergoes structural and functional remodeling during pregnancy. We hypothesize that myometrial genomic elements alter correspondingly in preparation for parturition. Human myometrial tissues from nonpregnant (NP) and term pregnant (TP) human subjects were subject to RNAseq, ATAC-seq and PGR ChIP-seq assays to profile transcriptome, assessible genome and PGR occupancy. Overall design: Three nonpregnant and three term pregnant myometrial specimens were subject to RNA-seq analysis. Viable tissues were taken from myometrial margins at the time of hysterotomy performed for clinically indicated cesarean section. After washing each specimen briefly in phosphate buffered saline, pieces of myometrium are preserved by plunging them into ice-cold RNALater. All specimens are stored at <-80°C until use.
