Two-Stage CAR T-Cell Differentiation in Patients with Large B-Cell Lymphoma

To investigate the CAR T-cell differentiation in vivo, we performed single-cell, multi-omics analyses of CD28-costimulated CAR T cells from patients with DLBCL who were successfully treated with axicabtagene ciloleucel. Our data shows that CAR T cells undergo two distinct stages of post-infusion clonal expansion. Data includes CAR T cells from infusion products and peripheral blood mononuclear cells from three patients with diffuse large B-cell lymphoma who achieved a complete response to axicabtagene ciloleucel. CAR T cells were sorted from peripheral blood using CD19-tetramers. Peripheral blood CAR T cells were analyzed at multiple timepoints within 90 days following administration of the infusion product to capture the CAR T-cell expansion, peak, and contraction phases.
