We have employed a fixed single nuclear RNA sequencing using the 10X Genomics FLEX assay to characterize the cellular constituents in healthy human scalp skin. Hair follicles are microanatomic niches that attract immune cells. Therefore, we generated this dataset to gain further insight into the chemokines kerotinocytes in skin express, and expression their cognate receptors on skin lymphocytes. Overall design: Two formalin-fixed paraffin-embedded healthy human scalp skin specimens were retrieved, and underwent tissue microtomy to generate three 50 micron thick tissue scrolls. The thick tissue scrolls underwent deparaffinization, rehydration, and mechanical and enzymatic dissocation using a Miltenyi GentleMacs Octodissociator with heaters. The resulting suspension was passed through a 30 micron strainer. Nuclei were counted using a Cellaca cell counter and submitted for standard 10X Genomics Flex assay.
