PC12 NGF Withdrawal and Stimulation Time Series

NGF (nerve growth factor) modulates pleiotropic functions including nervous system development and neuron excitability. PC12 cells are a well established model for studying NGF signaling in vitro. The objective of this study is to analyze the effects of NGF withdrawal and subsequent stimulation on gene transcription.PC12 cells were plated on collagen and cultured in low serum media with 50ng/ml NGF 72 hours (baseline). NGF was withdrawn from media for 48 hours, and subsequently replenished at 50ng/ml for 24 hours. Cultures were collected and RNA extracted for sequencing at baseline, after 8 hours and 48 hours of withdrawal, and after 8 hours and 24 hours of NGF replenishment.
