Human zinc finger protein 32 (ZNF32), localized in the nucleus, is a transcription factor involved in the development of breast cancer. Previous studies determined the nuclear localization sequence interval of ZNF32 and found that its histidine mutations at positions Aa179 and Aa183 (H179,183A) exhibited point-like nuclear speckles. However, the cause of ZNF32 nuclear speckles formation and its effects on breast cancer cells remain largely unknown. In this study, we successfully constructed ZNF32 histidine 179 and 183 double sites mutation (H179, 183A) and single site mutation (H179A, H183A) breast cancer cell lines, and we examined changes in genes associated with ZNF32 histidine 179 and183 single site mutation and double sites mutation in breast cancer cells by RNA-Seq.
