Multigenerational cell tracking of DNA replication and heritable DNA damage (scRNA)

The goal of this project was to analyze gene expression changes and gene expression heterogeneity at the single cell level upon DNA damage induced by ionizing radiation (IR). To this end, human U-2 OS cells were left untreated or exposed to 4 Gy of IR. 48 h after IR exposure, single cells were sorted into 384 well plates for scRNA-seq analysis. Overall design: For SORT-Seq, cells were sorted on a FACS Aria III 3L system. 48 hours prior to sorting, cells were treated with 4 Gy of ionizing radiation.
