Glucocorticoids unleash immune-dependent melanoma control through inhibition of the GARP/TGFβ axis [scRNA-seq]

Glucocorticoids were found to trigger immune-dependent control of an orthotopic melanoma model. We used scRNAseq to better understand the changes in tumour-infiltrating immune cells following glucocorticoid treatment. 10 mice were implanted with 20967 melanoma tumours intradermally. 5 mice were treated with control drug and 5 with glucocorticoid (clobetasol priopionate). These tumours were harvested, digested, pre-stained with anti-CD45 and HTO hashtags, and then the tumour-infiltrating immune cells were sorted, pooled, and subjected to scRNA sequencing.
