miRNA profile of extracellular vesicles isolated from bone marrow following in vivo irradiation

Male C57Bl/6 mice were irradiated with low and high dose X-ray irradiation. Mice were sacrificed 24 hours later and bone marrow-derived extracellular vesicles were isolated and microRNAs prepared. A miRNA profiling was performed by Exiqon (Vedbaek, Denmark), using qPCR panel with the miRNA Ready-to-Use PCR, Mouse&Rat panel I+II using ExiLENT SYBR® Green master mix. The amplification was performed in a LightCycler® 480 Real-Time PCR System (Roche) in 384 well plates The amplification curves were analysed using the Roche LC software, both for determination of quantification cycles (Cq) (by the 2nd derivative method) and for melting curve (Tm) analysis.
