Untargeted LC-MS/MS Metabolomics of Rabies-Infected Human Brain Tissues

DescriptionUntargeted LC-MS/MS metabolomics dataset of post-mortem human brain tissues from Rabies lyssavirus (RABV)-infected cases and age-matched controls across four anatomically distinct brain regions. Sample processing protocolPost-mortem brain tissues were obtained from eight laboratory-confirmed human rabies cases and eight control cases. Approximately 100 mg of frozen tissue from each sample was homogenized and metabolites were extracted using acetonitrile:methanol:water (2:2:1, v/v/v). Extracts from each brain region were pooled according to the experimental groups prior to LC-MS/MS analysis and data acquired in technical triplicates using a Vanquish UHPLC coupled to an Orbitrap Fusion Tribrid mass spectrometer in positive electrospray ionization mode. Data processing protocolRaw LC-MS/MS data were processed using Compound Discoverer 3.4 (Thermo Fisher Scientific) for peak detection, retention time alignment, feature grouping, and metabolite annotation. Quality control-based normalization was performed using the SERRF algorithm. Metabolites were annotated using the Human Metabolome Database (HMDB, version 5.0) within Compound Discoverer software. The dataset includes Thermo RAW files, and sample metadata.
