Nanoparticles formed at all PEGylation levels. Notably, the 80% NPs showed excellent stability, cytotoxicity, and cellular uptake but were also quickly cleared by the mononuclear phagocyte system (MPS) in vivo, causing SN38 leakage and intestinal toxicity. In comparison, the structural integrity of the 150% NPs was maintained in systemic circulation due to sufficient PEGylation, which allowed considerable accumulation at the tumor site through the enhanced permeability and retention (EPR) effect.
