Transcriptional profiling of ESCC treated with the selective HDACi LMK-235

Esophageal squamous cell carcinoma (ESCC) is an aggressive and deadly disease, which is the predominant subtype in China. Histone deacetylases (HDACs) and histone deacetylase inhibitors (HDACis) have been demonstrated that involved in ESCC carcinogenesis and anti-tumor activities, respectively. In this research, we deciphered the transcriptional landscape of KYSE150 treated with LMK-235, which was demonstrated the sensitive cell, and detected TNS3, one of the genes significantly down-regulated. TNS3 was distinctly over-expressed in ESCC, positively correlated with the malignant characteristics, and indicated poor prognosis in patients. Inhibition of TNS3 in ESCC cells inhibited proliferation both in vitro and in vivo, and sensitized the treatment responsive to LMK-235. In summary, we portrayed the LMK-235-treated transcriptome of ESCC cell, and detected the oncogene TNS3, which showed desensitization to the treatment of LMK-235.
