We performed 10x single cell multiome analysis (ATAC-seq + RNA-seq) on fetal wild-type and E47-/- N1End/d thymic cells to decipher the dynamics of chromatin structure and gene expression during thymic ontogeny. Live thymic cells were isolated from the fetal thymi of wild-type and E47-/- N1End/d mice at 15 days post coitum. These cells were then used to isolate nuclei and construct libraries for scRNA-seq and scATAC-seq from an identical nucleus, according to the manufacturer's protocol for Chromium Next GEM Single Cell Multiome ATAC + Gene Expression.
