Genomic regions containing active cis-regulatory DNA elements, which are bound with regulatory proteins, can be identified as DNase I hypersensitive sites (DHSs). In this study, we developed high-resolution DHS maps of potato (Solanum tuberosum) using chromatin isolated from leaves and tubers stored under room and cold (4°C) conditions, respectively. The dynamics of the chromatin accessibility as well as the changes in histone modifications of the cold-stressed potato tubers were analyzed. The regulations of differential gene expression in response to temperature variation versus tissue specificity were analyzed.
