Supplementary Materials for: Wood Smoke-Derived PM2.5 Exposure, Maternal Aortic Structure and Hemodynamic Adaptation in a Multigenerational Rat Model

This dataset contains six supplementary documents associated with a peer-reviewed manuscript reporting the effects of pregestational and gestational exposure to wood smoke-derived PM2.5 on maternal aortic structure and hemodynamic parameters in second-generation Sprague–Dawley rats. The study was conducted in Temuco, La Araucanía Region, Chile, during the austral winter of 2021, using a controlled environmental exposure system with parallel filtered (FA) and non-filtered air (NFA) chambers. Supplementary Document 1 provides aerial and ground-level photographs of the exposure chambers located in the courtyard of the Faculty of Medicine, Universidad de La Frontera, Temuco, along with geographic coordinates and proximity to the Las Encinas air quality monitoring station (500 m). Supplementary Document 2 presents descriptive statistics (mean ± SD and coefficient of variation) for ambient PM₂.₅, PM₁₀, and CO concentrations recorded during the study period (June 15 – September 30, 2021) and annually, obtained from the Las Encinas Monitoring Station of the National Air Quality Information System (SINCA). Supplementary Document 3 shows the daily time series of ambient PM2.5 concentrations (µg/m³) recorded at the Las Encinas Monitoring Station between January 1 and September 30, 2021, measured by a beta attenuation monitor BAM 1020 (Met One Instruments). Records are classified as validated, preliminary, and unvalidated. Supplementary Document 4 contains the complete morphometric dataset for the maternal aortic arch across four experimental groups (FA/FA, FA/NFA, NFA/FA, NFA/NFA; n = 12 per group), including thickness of the tunica intima, media, and adventitia; elastic fiber density; and inter-elastic fiber distance. Data are expressed as median and interquartile range, with Kruskal–Wallis and Dunn's post hoc statistics. Supplementary Document 5 presents maternal hemodynamic parameters — systolic blood pressure (SBP), diastolic blood pressure (DBP), mean arterial pressure (MAP), and heart rate (HR) — measured at gestational days 6, 12, and 18 post-fertilization using a non-invasive tail-cuff plethysmography system (CODA®, Kent Scientific) across all four experimental groups. Data are expressed as mean ± SD. Supplementary Document 6 provides the complete matrix of Spearman rank correlation coefficients and associated p-values between aortic wall thickness parameters (tunica intima, media, and adventitia) and hemodynamic variables (SBP, DBP, and HR at gestational day 18) for each experimental group. Supplementary Document 7 presents the Rate-Pressure Product (RPP) as an index of myocardial oxygen demand across gestational days 6, 12, and 18 post-fertilization. RPP was calculated as (Heart Rate × Systolic Blood Pressure) / 1000 for all four experimental groups (FA/FA, FA/NFA, NFA/FA, NFA/NFA; n = 12 per group). Data are expressed as mean ± SD. Group comparisons were performed using one-way ANOVA followed by Holm–Šídák post hoc test with FA/FA as reference. The document includes ANOVA p-values for each gestational day and demonstrates that RPP was significantly elevated in FA/NFA and NFA/NFA groups at GD12 and in FA/NFA and NFA/FA at GD18, indicating increased myocardial workload during mid-to-late gestation in animals exposed to unfiltered air during either exposure window.
