Ronin knockout is embryonic lethal and Ronin knockout ES cells are not viable. Here we used CreER2; Roninflox/flox mouse ES cells to induce Ronin knockout by tamoxfien treatment in comparison to control Roninflox/flox cells to study Ronin knockout related transcriptional changes. Roninflox/flox (control) or CreERT2; Roninflox/flox mouse ES cells were plated at a density of 100,000 cell per 10 cm2 and cultured in standard mouse ES cell medium. Cells were treated with 0.25 µM tamoxifen for 1.5, 3 or 6 days and total RNA was isolated to study gene expression changes.
