Characterization of the gene regulatory cascade downstream of master-regulator STZ

STZ acts as a brige between PEP1-induced transcription factor and the PEP1-suppressed cell-cycle genes. In the conditional STZ overexpression seedlings (p35S::XVE>>pLexA::STZ), we observed an early differentiation of tissues. To identify the genetic cascade regulated by STZ, we prepare samples at 6h and 24h-post estradiol induction. The total RNA ws extracted and bulk RNA sequiencing was performed. Overall design: We took 4-5 days old Arabidopsis seedling and treated them with 20 µM estradiol for 6h and 24h. Following STZ-induction, we dissected the apical 2/3 portion of the root and the RNA was extrcted using RNeasy Plant Mini kit (Qiagen). The purification of the samples and sequencing was done by Macrogen (Korea).
