Here we developed “GradingPoolSeq”, a new approach combining high-throughput sequencing with bulked-segregant analysis (BSA), for QTL mapping in crop breeding. We could rapidly detect multiple QTLs of the target trait simultaneously by only requiring the second generations, reducing a large amount of time needed in constructing recombinant inbred lines by applying traditional QTL analysis.
