The objective of this project is to investigate the role of the R2TP complex in paramyxovirus infection. Our data has demonstrated that the R2TP complex interacted with the paramyxovirus polymerase L protein and regulated viral RNA synthesis. To clarify precise mechanism of viral RNA synthesis, we analyzed viral and host gene expressions by RNA deep sequencing. For mumps virus (MuV), viral gene transcription, but not genome replication, was uncontrollably upregulated, when the R2TP complex expression was silenced. This regulation by the R2TP complex was critical for MuV to ensure its replication by evading the host innate immune responses. The R2TP complex also functions as a regulator/suppressor of measles virus (MeV) RNA synthesis, but the function was simply inhibitory.
