Cigarette smoke induced macrophage senescence

This project profiles cigarette smoke-associated macrophage dysfunction and senescence using two complementary datasets. First, single-cell RNA-seq (10x Genomics) was performed on bronchoalveolar lavage fluid (BALF) cells collected from human participants with different smoking histories (never, former, current). Second, bulk RNA-seq was performed on PMA-differentiated THP1 cells (THP1-M) exposed to 5% cigarette smoke extract (CSE) for 24 h versus controls. Libraries were sequenced on Illumina NovaSeq 6000. The study aims to define cell-state changes and identify regulators (e.g., GEM) and pathways (e.g., JAK/STAT3) underlying cigarette smoke-driven macrophage senescence.
