Aim: Analyse tumour mutations related to ethanol consumption Wild type and Aldh1b1 hypomorph mice received 20% ethanol in drinking water or normal drinking water for one year. 4 of 14 ethanol-treated wild type mice and 4 of 15 ethanol-treated Aldh1b1 knockout mice developed intestinal tumours (8 tumours in total in wt and 11 in hypomorph group) while none of the drinking water controls (n=15 per genotype) developed any intestinal tumours. Tumour DNA will be isolated from FFPE tumour samples. (Expecting 6 wt and 8 hypomorph tumours to yield enough material.) To increase sample numbers, APCmin mice received 20% ethanol in drinking water or normal drinking water for 9-10 weeks and DNA was isolated from tumours that were collected from the freshly dissected intestine and snap frozen. (DNA has been isolated from 50 tumours per group, i.e. 100 samples in total.)
